If there is not a solid connection between the pads and your pin/wires, you will have issues uploading. Try checking your connections and making sure your solder joints are ideal. It's probably that the you did not add a jumper from the DTR pin to reupload correctly. It’s tricky but as you you reset the board, it will be in bootloader mode so that you can upload code before it starts running the code that was uploaded.
